ABSTRACT Fungi of Pleurotus genus have attracted a great interest due to their medicinal properties such as anti-inflammatory, antimicrobial and antitumor. These properties are attributed mainly to polysaccharides synthesized by Pleurotus. This work aimed to study the mycelial growth of P. ostreatus in submerged culture, evaluating the influence of the initial concentration of substrate (20 and 40 g/L of glucose) and the pH (4 and 6) on kinetic parameters of production of biomass. Aflatoxin detoxification by manganese peroxidase purified from Pleurotus ostreatus BJM

Manganese peroxidase (MnP) was produced from white rot edible mushroom Pleurotus ostreatus on the culture filtrate. The enzyme was purified to homogeneity using (NH4)2SO4 precipitation, DEAE-Sepharose and Sephadex G-100 column chromatography. The final enzyme activity achieved 81UmL-1, specific activity 78 U mg-1 with purification fold of 130 and recovery 1.2% of the crude enzyme. SDS-PAGE indicated that the pure enzyme have a molecular mass of approximately 42 kDa. The optimum pH was between 4-5 and the optimum temperature was 25 ºC. The pure MnP activity was enhanced by Mn2+,Cu2+,Ca2+ and K+ and inhibited by Hg+2 and Cd+2.H2O2 at 5 mM enhanced MnP activity while at 10 mM inhibited it significantly. Applicability of the use of waste from different banana cultivars for the cultivation of the oyster mushroom BJM

The objective of this research was to evaluate the oyster mushroom Pleurotus ostreatus- (Jacq.: Fr.) Kumm. cultivation in substrates based on different combinations of wastes (leaf, pseudo-stem and pseudo-stem + leaf) and banana cultivars - Musa spp. (Thap Maeo, Prata Anã, Pelipita and Caipira) during 49 days. Organic matter loss in the substrate by action of the fungus was also evaluated during that period. It was verified that the pseudo-stem waste provided the best averages of biological efficiency among all cultivars tested and best rates were obtained by Thap Maeo (61.5%). En la producción de hongos comestibles se utilizan diversos residuos agroindustriales como la paja de cebada, trigo y avena. Como resultado del cultivo, se producen grandes cantidades de sustrato residual, el cual puede potencialmente ser usado para procesos de biorremediación in situ. En el presente estudio, se cultivó el hongo comestible Pleurotus ostreatus (CP-50) en presencia del hidrocarburo aromático policíclico, antraceno. La cepa se cultivó en tres condiciones: 1) Agar con Extracto de Malta (EMA), CP-50 y antraceno (tratamiento); 2) EMA y CP-50 (testigo) y 3) EMA y antraceno (blanco). Cultivo e características nutricionais de Pleurotus em substrato pasteurizado Bragantia

O objetivo deste trabalho foi avaliar a produtividade, eficiência biológica, massa fresca, composição centesimal dos cogumelos Pleurotus ostreatus (BF24) e Pleurotus sajor-caju (PSC96/03 e PSC01/06) produzidos no substrato capim-elefante (Pennisetum purpureum) pasteurizado e a relação Carbono/Nitrogênio inicial e final do substrato. O substrato seco e particulado a 2 cm foi umedecido por 24 horas e pasteurizado a 100 ºC durante 30 minutos. Adicionaram-se 3% de inóculo de cada linhagem, sendo acondicionado em embalagens de polipropileno com 1 kg cada uma. Os substratos foram incubados a 26 ºC e na fase de frutificação a 23±3 ºC e umidade relativa de 75% a 90%. La producción de “setas” (Pleurotus ostreatus), es una alternativa para producir y diversificar los alimentos que requiere la población en México. En el país es escasa la información acerca de la rentabilidad de éste cultivo. Por ello, el objetivo de este trabajo fue estimar una función de producción que muestre matemáticamente la relación funcional entre los insumos productivos utilizados y la producción “setas”. La variable dependiente la constituyó la producción de “setas” producidas y la variable independiente fueron las bolsas en producción. Drying and rehydration of oyster mushroom BABT

Dehydration and rehydration processes of Pleurotus ostreatus fruiting bodies were investigated in this work. Mushroom samples were dehydrated at 40, 50 and 60 ºC, using drying air with relative humidity of 75 %. The rehydration was investigated at different temperatures of immersion water (25, 55 and 85 ºC) and different immersion times (30, 75 and 120 minutes). The best rehydration occurred for the samples dried at 40 ºC. The rehydration could be done in water at room temperature, during 30 minutes. Water sorption isotherms of samples were determined at 30, 40 and 50 ºC. Both GAB and BET models satisfactorily represented the experimental data of moisture sorption of dried mushrooms.

Trichoderma spp is the cause of the green mold disease in mushroom cultivation production. Many disinfection treatments are commonly applied to lignocellulose substrates to prevent contamination. Influence of initial pH of the growing medium on the activity, production and genes expression profiles of laccase of Pleurotus ostreatus in submerged fermentation Electron. J. Biotechnol.

Background: Enzymatic activity and laccase isoenzymes number of Pleurotus ostreatus grown in different pH values of the growing medium in submerged fermentation and incubated in buffer solutions of different initial pH values were determined. The expression profiles of five laccase genes (Lacc1, Lacc4, Lacc6, Lacc9 and Lacc10) in these cultures were also studied. Results: The highest laccases activity was obtained in cultures grown at initial pH of 4.5 and the lowest in cultures grown at initial pH of 8.5. Isoenzyme profiles were different in all the cases. Mycelial growth of strains of Pleurotus ostreatus developed on agar and its correlation with the productivity in pilot production farm BJM

Radial growth rate, intracellular laccases and proteases activities, and protein content were evaluated in five strains of Pleurotus ostreatus, grown on starch-based and glucose-based agar media containing different concentrations of the glucose analogue 2-deoxyglucose (2-DG). Productivity of the strains in pilot scale cultivation was also determined. The mycelium of four strains had approximately between 0.6- to 3-fold higher protein content when grown on glucose medium containing 0.01 g/L of 2-DG than when grown on glucose medium. The radial growth rate and intracellular laccases activity of some strains showed a positive and a negative correlation with the productivity, respectively. The substrate designed for mushroom cultivation is achieved through a controlled composting process from agricultural by-products (chicken manure and wheat straw). This work shows the potential of DAM as efficient compost supplement for culturing the species Agaricus bisporus (J.E. Lange) Imbach and Pleurotus ostreatus (Jacq.) P. Kumm. Supplementation during A. bisporus cultivation results in larger mushrooms with a firmer texture and higher dry matter and protein contents in comparing with the non-supplemented substrate. Abstract Polysaccharides from fungal wastes were partially characterized and evaluated for their protective effects against bacterial spot caused by Xanthomonas gardneri on four tomato cultivars: Santa Cruz Kada, Natália, BRS Sena and Forty. The polysaccharides were extracted from spent mushroom substrate of Pleurotus ostreatus, residual brewery yeast (Saccharomyces cerevisiae), and basidiocarps discarded from Lentinula edodes production. Use of remazol blue dyed avicel for the determination of cellulolytic activity in basidiomycetes Rev. Microbiol.

A modified method for direct determination of cellulolytic activity using Avicel colored with Remazol Brilliant Blue R (RBBR) in Agar test tubes is discussed. Refinements were introduced in a simple method for quantitation of cellulase activity, based on the release of dye from Avicel-RBBR medium by the enzymatic hydrolysis. Modifications in Avicel-dye preparation were enhanced and a spectrophotometer for direct OD measurement in agar test tubes used. The use of a spectrophotometer improved the precision of the collected data, since absorbance measurements could be done at the maximum wavelenght for RBBR (595 nm).
